RESUMO

Schizophrenia (SZ) is a neurodevelopmental-associated disorder strongly related to environmental factors, such as hypoxia. Because there is no cure for SZ or any pharmacological approach that could revert hypoxia-induced cellular damages, we evaluated whether modulators of sirtuins could abrogate hypoxia-induced mitochondrial deregulation as a neuroprotective strategy. Firstly, astrocytes from control (Wistar) and Spontaneously Hypertensive Rats (SHR), a model of both SZ and neonatal hypoxia, were submitted to chemical hypoxia. Then, cells were exposed to different concentrations of Nicotinamide (NAM), Resveratrol (Resv), and Sirtinol (Sir) for 48hrs. Our data indicate that sirtuins modulation reduces cell death increasing the acetylation of histone 3. This outcome is related to the rescue of loss of mitochondrial membrane potential, changes in mitochondrial calcium buffering capacity, decreased O2-rad levels and increased expression of metabolic regulators (Nrf-1 and Nfe2l2) and mitochondrial content. Such findings are relevant not only for hypoxia-associated conditions, named pre-eclampsia but also for SZ since prenatal hypoxia is a relevant environmental factor related to this burdensome neuropsychiatric disorder.

BACKGROUND: Relative age effect (RAE) is a concept related to the possible advantage that older athletes would have over younger ones within the same category. Although many studies have approached this subject in individual sports, there are few clippings by events within the sport. More detailed analyses are necessary for a better understanding of how RAE behaves in sports, especially in athletics, the subject of this study. The objective of this study was to analyze the RAE on speed in track and field events as a whole, separating the flat races from the hurdles races. METHODS: The Brazilian Ranking of Brazilian Athletics Confederation was used for data analysis, and the sample was composed of the 50 best-placed marks in the ranking of speed events in athletics in the categories Under(U)-16 and U-18 (female and male). Statistical analysis was calculated by chi-square, and the effect size was checked by Cramer's V. Likelihood-ratio test (L-Ratio) assessed the probability of the RAE occurring in the total sample and by age groups. RESULTS: In the total sample the results pointed to the emergence of RAE in males in both categories (U-16: p < 0.001; V: 0.13; L-Ratio: 3.64, U-18: p < 0.001; V: 0.13; L-Ratio: 3.80), whereas in females no such effect was found in any category (U-16: p = 0.6; V: 0.09; L-Ratio: 0.09, U-18: p = 0.6; V: 0.07; L-Ratio: 0.12). When the results were separated by type of event, there was only a RAE in the shallow event in the U-18 female category (p = 0.3; V: 0.11; L-Ratio: 8.72). CONCLUSION: The results allow us to conclude that there is a RAE in the speed trials of Brazilian athletics in the U16 and U18 categories for men, while this effect appears only in the shallow trials of the U18 category for women, indicating that the RAE has incidence when there is more participation and competition in the sport.

ABSTRACT BACKGROUND: Presenteeism refers to the presence of a worker at work with reduced performance due to illness, and it is a common public health problem. Exposure to noise during production processes brings risk to workers' health. OBJECTIVE: To analyze the health profile of workers in the automotive industry and identify the association between noise perception and presenteeism among workers in the Brazilian automotive industry. DESIGN AND SETTING: This was a cross-sectional study utilizing a case study design analyzing the automotive industry in the interior of São Paulo state, Brazil. METHODS: This study included 306 workers using the Presenteeism Work Limitations Questionnaire protocol. Multiple logistic regression was used for data analysis. RESULTS: Male workers with reports of headache, tension, and limited well-being at work, having perceived that noise exposure interferes with productivity, showed a positive association with the occurrence of presenteeism. Physical demand had the highest score in terms of interfering with the presenteeism index. CONCLUSION: Workers' perceptions of noise were associated with presenteeism.

Background: Researches are linking Biological Maturation (BM) with the performance of adolescent rowers from both genders. Despite this, there is still not enough information about the influence of BM advancement correlating to sports, aerobic and anaerobic performance in adolescent athletes at the sport modality rowing. Objective: Investigate the influence of Biological Maturation on sports performance and aerobic and anaerobic power in adolescent rowing athletes. Methods: A longitudinal observational study, developed over 3 years, with a sample of 52 adolescents, rowing athletes, of both genders (61% male and 39% female) mean age of 16.0 ± 0.5 years old at the start and 18.4 ± 0.5 years old at the end of the study. Analysis was performed once a year. BM was evaluated through maturational groups generated from Age Peak Height Velocity; maximum aerobic power [VO2Max (ml/kg/min)] and mean anaerobic power (Watts) through the ergometer test (indoor rowing); peak anaerobic power (Watts) through a mathematical model derived from competition time, to determine sports performance analyzed the race time during world championship tryouts. Results: The advancement of BM influenced the reduction of the test time and increase of the mean anaerobic power (Watts) in indoor rowing (η2p > 0.36, p < 0.05), as well as an improvement in performance in sports competition (η2p > 0.35, p < 0.05). However, the advancement of BM did not affect VO2Max (ml/kg/min) in young elite rowing athletes of both sexes (p > 0.05). Conclusion: Advances in biological maturation have been shown to influence the anaerobic and sports performance (reduction of the execution time in 2,000-m) of adolescent rowing athletes of both genders.

Trypanosoma cruzi requires a triatomine insect vector for its life cycle, which can be complex in different enzootic scenarios, one of which is the unique transmission network in the Atlantic Forest of south-eastern Brazil. In Espírito Santo (ES) State, highly infected Triatoma vitticeps are frequently reported invading domiciles. However, triatomines were not found colonizing residences and mammals in the surrounding areas did not present T. cruzi infection. To date, the biotic and abiotic variables that modulate T. vitticeps occurrence and T. cruzi infection in ES State are still unknown. The aim of this study was to identify the environmental variables that modulate their occurrence. Local thematic maps were generated for two response variables: T. vitticeps occurrence and T. cruzi infection. The following explanatory variables were tested: climate (temperature, relative air humidity and rainfall), altitude elevation, mammalian species richness as well as soil and vegetation types. Spatiotemporal distribution patterns and correlation levels between response and explanatory variables were assessed through spatial statistics and map algebra modelling. The central and southern mesoregions presented higher T. vitticeps and T. cruzi distributions and can be considered transmission hotspots. The explanatory variables that can explain these phenomena were relative air humidity, average temperature, soil type, altitude elevation and mammalian species richness. Algebra map modelling demonstrated that central and southern mesoregions presented the environmental conditions needed for T. vitticeps occurrence and T. cruzi infection. The consideration of environmental variables is essential for understanding the T. cruzi transmission cycle. Cartographic and statistical methodologies used in parasitology have been demonstrated to be reliable and enlightening tools that should be incorporated routinely to expand the understanding of vector-borne parasite transmission.

BACKGROUND: The exhaustive series of tests undergone by young athletes of Olympic rowing prior to important competitions imply loads of physical stress that can ultimately impact on mood and motivation, with negative consequences for their training and performance. Thus, it is necessary to develop a tool that uses only the performance of short distances but is highly predictive, offering a time expectancy with high reliability. Such a test must use variables that are easy to collect with high practical applicability in the daily routine of coaches. OBJECTIVE: The objective of the present study was to develop a mathematical model capable of predicting 2000 m rowing performance from a maximum effort 100 m indoor rowing ergometer (IRE) test in young rowers. METHODS: The sample consisted of 12 male rowing athletes in the junior category (15.9 ± 1.0 years). A 100 m time trial was performed on the IRE, followed by a 2000 m time trial 24-h later. RESULTS: The 2000 m mathematical model to predict performance in minutes based on the maximum 100 m test demonstrated a high correlation (r = 0.734; p = 0.006), strong reliability index (ICC: 0.978; IC95%: [0.960; 0.980]; p = 0.001) and was within usable agreement limits (Bland -Altman Agreement: -0.60 to 0.60; 95% CI [-0.65; 0.67]). CONCLUSION: The mathematical model developed to predict 2000 m performance is effective and has a statistically significant reliability index while being easy to implement with low cost.

ABSTRACT: The physiological benefits of applying blood flow restriction (BFR) in isolation or in the presence of physical exercise have been widely documented in the scientific literature. Most investigations carried out under controlled laboratory conditions have found the technique to be safe. However, few studies have analyzed the use of the technique in clinical settings.To analyze how the BFR technique has been applied by professionals working in the clinical area and the prevalence of side effects (SEs) resulting from the use of this technique.This is a cross-sectional study. A total of 136 Brazilian professionals who perform some function related to physical rehabilitation, sports science, or physical conditioning participated in this study. Participants answered a self-administered online questionnaire consisting of 21 questions related to the professional profile and methodological aspects and SEs of the BFR technique.Professionals reported applying the BFR technique on individuals from different age groups from youth (≤18 years; 3.5%) to older adults (60-80 years; 30.7%), but mainly on people within the age group of 20 to 29 years (74.6%). A total of 99.1% of the professionals coupled the BFR technique with resistance exercise. Their main goals were muscle hypertrophy and physical rehabilitation. The majority (60.9%) of interviewees reported using BFR in durations of less than 5 minutes and the pressure used was mainly determined through the values of brachial blood pressure and arterial occlusion. Moreover, 92% of professionals declared observing at least 1 SE resulting from the BFR technique. Most professionals observed tingling (71.2%) and delayed onset of muscle soreness (55.8%). Rhabdomyolysis, fainting, and subcutaneous hemorrhaging were reported less frequently (1.9%, 3.8%, and 4.8%, respectively).Our findings indicate that the prescription of blood flow restriction technique results in minimal serious side effects when it is done in a proper clinical environment and follows the proposed recommendations found in relevant scientific literature.

Depletion of oxygen (O2) levels and reduction in the ATP synthesis (or even its complete blockage) are important characteristics of mitochondrial dysfunction; features that are often correlated with neurodegeneration. The measurement of oxygen consumption rate (OCR) is thus essential to evaluate cellular metabolism, survival, and neuroprotective strategies. In the present chapter, we describe the oxygen consumption assay using a Clark-type oxygen electrode in different types of samples named cells suspension (from primary and established cell culture), brain slices (ex vivo), and fresh brain tissues. In addition, we demonstrate herein how the program Oxygraph can be used in order to analyze the data and different approaches to normalize it.

One of the highest incidences of illegal drug products is related to phosphodiesterase-5 inhibitors, used in treatment of erectile dysfunction, including those containing sildenafil citrate and tadalafil. In this context, comprehensive evaluation of the quality of genuine and illegal medicines was performed. A simple and rapid ultra-high performance liquid chromatography (UHPLC-UV) method to quantify sildenafil and tadalafil in the presence of six degradation products was developed and validated. Sildenafil and tadalafil were submitted to forced degradation. The separation was carried out on a Kinetex C18 (50 × 2.1 mm; 1.7 µm) column with mobile phase composed of acetonitrile and aqueous triethylamine solution. The calibration curves were linear in the range of 14-126 µg mL-1 for sildenafil citrate and 4-36 µg mL-1 for tadalafil and the method proved to be selective, precise, accurate and robust. Sildenafil degraded in oxidative media, whereas tadalafil degraded in acidic, alkaline and oxidative environment. The chemical structures and the mechanisms for the formation of the main degradation products were proposed by UHPLC coupled to tandem mass spectrometry. The UHPLC-UV method was applied in the pharmaceutical analysis of genuine and seized medicines. Some of them did not meet quality standards, mainly due to contents below specifications and the large variation on contents between units within a batch.

The present study aimed to determine the effect of mental fatigue (MF) on total training volume (TTV; number of repetitions x number of sets x load) and on ratings of perceived exertion (RPE; Borg, 1982) in the half-back squat exercise (HBSE). Nine male subjects (M age = 22.6 years, SD = 2.3; M height = 172.3 cm, SD = 6.8; M weight = 76.2 kg, SD = 9.8; M years of resistance training experience = 4.1, SD = 2.3 years) recruited from a university population were study participants in this participant-blind cross-over and randomized study. Participants underwent either the Stroop task - a highly demanding cognitive task (CT) - or a control condition (CON) in which they viewed a documentary exhibition for 30 minutes. Perception of MF and motivation were assessed after treatments using a visual analog scale of 100 mm. Participants then engaged in a countermovement jump (CMJ) test and three sets of HBSE until they reached momentary concentric failure, reporting RPE at the end of each exercise set. Following the CT, participants showed a significantly increased self-perception of MF in relation to the CON condition (p = 0.01; d = 1.2), but this did not affect their motivation to engage in subsequent tests (p = 0.99; d = 0.006). Neither the CMJ performances nor the RPE were statistically different between CT and CON conditions (p = 0.33; d = 0.09 and p = 0.20; η2 = 0.20, respectively). TTV was significantly lower in the CT relative to the CON experimental condition (Δ = -15.8%; p = 0.04; η2 = 0.48). Prolonged involvement in a CT was associated with reduced volume on a resistance exercise, though this effect was not associated with changes in CMJ performance or motivation to exercise.

PURPOSE: To develop an equation to provide the peak power (PP) through a specific stimulus performed in an aquatic environment, as well as to correlate morphological, anthropometric and strength variables with rowing performance. METHODS: The sample consisted of 16 elite young rowing athletes of both sexes (15.7 ± 1.21 years). The strength of upper limbs and lower limbs was verified. To analyze the PP, a 100-m Sprint test was performed on an indoor rowing type ergometer, and after a 72-hour wash-out, the test was repeated in an aquatic environment on a vessel equipped with a global position system. Body composition was analyzed by examining bone densitometry with an X-ray source and maturation was verified by anthropometry. RESULTS: The tests for water sprint and indoor rowing showed significant reliability (ICC = 0.695; p = 0.0007). The PP aquatic showed reliability with that acquired in indoor rowing (ICC = 0.897; p<0.0001) and was related to maturation (p<0.05). The morphology, anthropometry and strength of the upper limbs were related to the sprint and peak strength in both tests (p<0.05). CONCLUSION: The equation for the PP in aquatic environment presented by the present study is highly reliable with an indoor ergometer digital ergometer.

The present study was conducted to evaluate the socioeconomic inequality related to lung cancer mortality rates and trends between 2000 and 2015 according to gender in Brazil. We retrieved the death records for cases of lung cancer (ICD-10 C33 C34) from 2000 to 2015 in adults age 30 years and older in Brazilian Regions from official databases (DATASUS) and corrected for ill-defined causes. The Prais-Winsten regression method and Pearson correlation were applied. The results were considered statistically significant when p < 0.05. The correlation between the lung cancer mortality rates and the HDI decreased when the rates for the first and last years of the historical series were compared in men (r = 0.77; r = 0.58) and women (r = 0.64; r = 0.41). However, the correlation between the trends in the lung cancer mortality rates and the HDI was negative in men (r = - 0.76) and women (r = - 0.58), indicating larger reductions (or smaller additions) among the Federative Units with the highest HDI, in contrast to trends reflecting a greater increase in those with the lowest HDI. Our results suggest a relevant inequality in the trends of mortality from lung cancer in Brazil.

O ruído ocupacional representa um risco à saúde dos trabalhadores, como perda auditiva e zumbido, ainda muito predominante em diversos ambientes e processos. O presenteísmo é definido como um fenômeno em que o trabalhador se encontra fisicamente no trabalho, mas por diversos fatores tem sua concentração e dedicação prejudicadas na realização da atividade. Este estudo tem como objetivo identificar que fatores estão associados, incluindo aqueles relacionados à exposição ao ruído, à ocorrência de presenteísmo. O estudo se caracteriza como exploratório, com abordagem quantitativa. No delineamento foi realizado um estudo de caso em uma empresa localizada no Vale do Paraíba, com uma amostra de 23 trabalhadores da indústria da mineração. Os dados foram obtidos por meio da aplicação do protocolo Work Limitations Questionnaire de presenteísmo e de questionário de dados sociodemográficos. A análise de associação dos desfechos foi realizada por meio de regressão logística múltipla. Foi possível verificar que, a demanda física foi a que teve maior escore, bem como o fator zumbido pode ser considerado como variável que influencia o presenteísmo.


Occupational noise represents a risk to workers' health, such as hearing loss and tinnitus, still very prevalent in various environments and processes. Presenteeism is defined as a phenomenon in which the worker is physically at work, but due to various factors, his concentration and dedication are impaired in performing the activity. This study aims to identify which factors are associated, including those related to noise exposure, to the occurrence of presenteeism. The study is characterized as exploratory, with quantitative approach. In the design a case study was carried out in a company located in the Vale do Paraíba, with a sample of 23 workers from the mining industry. Data were obtained by applying the Work Limitations Questionnaire presenteeism protocol and the sociodemographic data questionnaire. The outcome association analysis was performed by multiple logistic regression. It was possible to verify that the physical demand was the one with the highest score, and the tinnitus factor can be considered as a variable that influences the presenteeism.


El ruido laboral representa un riesgo para la salud de los trabajadores, como la pérdida de audición y el tinnitus, que sigue siendo muy frecuente en diversos entornos y procesos. El presentismo se define como un fenómeno en el que el trabajador está físicamente en el trabajo, pero debido a varios factores, su concentración y dedicación se ven perjudicados para realizar la actividad. Este estudio tiene como objetivo identificar qué factores están asociados, incluidos los relacionados con la exposición al ruido, a la ocurrencia de presentismo. El estudio se caracteriza por ser exploratorio, con enfoque cuantitativo. En el diseño se realizó un estudio de caso en una empresa ubicada en Vale de Paraíba, con una muestra de 23 trabajadores de la industria minera. Los datos se obtuvieron aplicando el protocolo de presentismo WLQ y el cuestionario de datos sociodemográficos. El análisis de asociación de resultados se realizó mediante regresión logística múltiple. Fue posible verificar que la demanda física fue la que obtuvo la puntuación más alta, y el factor tinnitus puede considerarse como una variable que influye en el presentismo.

Amyotrophic lateral sclerosis (ALS) is a multifactorial and progressive neurodegenerative disease of unknown etiology. Due to ALS's unpredictable onset and progression rate, the search for biomarkers that allow the detection and tracking of its development and therapeutic efficacy would be of significant medical value. Considering that alterations of energy supply are one of ALS's main hallmarks and that a correlation has been established between gene expression in human brain tissue and peripheral blood mononuclear cells (PBMCs), the present work investigates whether changes in mitochondrial function could be used to monitor ALS. To achieve this goal, PBMCs from ALS patients and control subjects were used; blood sampling is a quite non-invasive method and is cost-effective. Different parameters were evaluated, namely cytosolic calcium levels, mitochondrial membrane potential, oxidative stress, and metabolic compounds levels, as well as mitochondrial dynamics and degradation. Altogether, we observed lower mitochondrial calcium uptake/retention, mitochondria depolarization, and redox homeostasis deregulation, in addition to a decrease in critical metabolic genes, a diminishment in mitochondrial biogenesis, and an augmentation in mitochondrial fission and autophagy-related gene expression. All of these changes can contribute to the decreased ATP and pyruvate levels observed in ALS PBMCs. Our data indicate that PBMCs from ALS patients show a significant mitochondrial dysfunction, resembling several findings from ALS' neural cells/models, which could be exploited as a powerful tool in ALS research. Our findings can also guide future studies on new pharmacological interventions for ALS since assessments of brain samples are challenging and represent a relevant limited strategy. BACKGROUND: Endocrine mechanisms can be a determining factor in the neuromuscular performance of young athletes. OBJECTIVE: The objective of the present study was to relate maturational and hormonal markers to neuromuscular performance, as well as to verify whether young athletes with different testosterone levels show differences in muscle strength. METHODS: The sample consisted of 37 young male Brazilian athletes (11.3 ± 0.94 years) who were members of a sports initiation project. Hormonal markers were analyzed biochemically by blood samples, and maturation markers by mathematical models based on anthropometry. Body composition was verified by tetrapolar bioimpedance. The performance of upper and lower limb strength and body speed were analyzed. RESULTS: Hormonal and maturational markers were related to neuromuscular performance (p < 0.05). Young people with higher testosterone levels showed higher muscle strength (p < 0.05). Artificial neural networks showed that testosterone predicted the performance of upper limbs by 49%, and maturation by 60%. Maturation foreshadowed the performance of lower limbs by 30.3%. CONCLUSION: Biological maturation and hormonal levels can be related to neuromuscular performance, and young people with higher testosterone levels show superior muscle strength in relation to the others.

BACKGROUND: Occupational accidents are a complex phenomenon and a major public health problem. Occupational health surveillance actions are essential for prevention of injuries of this nature. OBJECTIVE: To ascertain the prevalence of and the variables associated with occupational accidents in the city of Itajubá (MG). DESIGN AND SETTING: A cross-sectional study with a quantitative approach, based on a household survey with random sampling, was conducted in areas covered by the Family Health Strategy (FHS) in Itajubá (MG). METHODS: Questionnaires were applied to 292 people. The data were analyzed by means of logistic regression. RESULTS: The prevalence of occupational accidents was 8.6%. The underreporting rate was 60.0%. The scenario for these accidents, according to the model established through the regression analysis, was most likely to involve males who declared their skin color as white and who did not have a formal employment contract. CONCLUSION: This study makes a contribution towards unveiling the relationship between healthcare and work, and thus serve as support for the development of strategies to prevent underreporting. Lastly, the results provide the basis for future public health intervention actions and for future studies.

ABSTRACT Purpose: investigate the association of noise exposure with the occurrence of systemic arterial hypertension (SAH) among public school female teachers. Methods: a cross-sectional, descriptive and observational study was conducted. A questionnaire elaborated by the researchers was applied to a random sample of 133 female teachers. Noise exposure dosimetry was used among the sample subjects. The non-conditional logistic regression technique was applied to identify significant variables and control confounding variables for the occurrence of SAH. Results: the prevalence of hypertension found, in teachers, in this study, was 17.3%. However, the association between occupational noise exposure and the occurrence of hypertension was not verified. It was observed that smoking is a very relevant risk factor for hypertension, while the physical effort performed at work appeared as a protection factor. Conclusion: although the variable "noise exposure dose" was not significant in the final model, it was found that the exposure values were above the limits, justifying the adoption of preventive measures.

ABSTRACT This study ivestigated the association between noise and impaired sleep quality in 150 physical education instructors, in bodybuilding gyms. Two questionnaires were applied: evaluation of subjective sleep quality and analysis of explanatory variables. Noise exposure was analyzed. For the data analysis, was used the technique of non-conditional logistic regression, to obtain the values of the adjusted odds ratios (OR) of the significant variables. Two proper adjustment models stood out. The first model presented the significant variables: incorrect performance of the meals (OR = 2.58, 95% CI 1.08 to 6.02); skin color - black ou brown (OR = 0.53, 95% CI 0.25 to 1.12) and exposure to noise level above or equal to 80 dB(A) (OR = 1.90; 95% CI 0.87 to 4.12). The second model presented the significant variables: incorrect performance of the meals (OR = 2.35, 95% CI 0.1 to 5.56); exposure level standardized above or equal to 80 dB(A) (OR = 2.03, 95% CI 0.92 to 4.47) and use 0of drugs for depression/anxiety (OR = 0.26; 95% CI 0.53 to 1.33). This research identified the variables that were significantly associated with impaired sleep quality, as exposure to noise, contributing to the adoption of preventive measures.


RESUMO Este estudo investigou a associação entre ruído e qualidade prejudicada do sono em 150 instrutores de educação física, em academias de musculação. Dois questionários foram aplicados: avaliação da qualidade subjetiva do sono e sobre variáveis explanatórias. A exposição ao ruído foi avaliada. Para a análise dos dados, utilizou-se a técnica de regressão logística não-condicional, para obter os valores das Razões de Chances (RC) das variáveis significativas. Dois modelos finais se destacaram. O primeiro apresentou as variáveis significativas: realização incorreta das refeições (RC = 2,58, IC 95% 1,08 a 6,02); cor da pele - preta ou parda (RC = 0,53; IC 95% 0,25 a 1,12) e exposição ao ruído acima ou igual a 80 dB(A) (RC = 1,90; IC95% 0,87 a 4,12). O segundo modelo apresentou as variáveis significativas: realização incorreta das refeições (RC = 2,35, IC 95% 0,1 a 5,56); nível de exposição padronizado acima ou igual a 80 dB (A) (RC = 2,03, IC 95% 0,92 a 4,47) e uso de medicamentos para depressão/ansiedade (RC = 0,26; IC95% 0,53 a 1,33). Esta pesquisa identificou as variáveis que se associaram significativamente com a qualidade do sono prejudicada, como a exposição ao ruído, contribuindo para a adoção de medidas preventivas.
